A pre-concentration and on-line solid phase extraction setup for the LC-MS analysis of therapeutic protein mixtures

Instrument Type: LCMS

This study shows a fully automated UHPLC setup applying a simple and efficient online solid phase extraction method. Showcase assay performance by using a column cartridge for sample trapping and a Thermo Scientific™ MAbPac™ RP column for separation with water/acetonitrile-based gradients. Leverage the new technologies such as the Thermo Scientific™ Vanquish™ UHPLC platform in combination with the Thermo Scientific™ Q Exactive™ HF Hybrid Quadrupole Orbitrap mass spectrometer.

Quantitative Forensic Analysis of Opiates, Opioids, and Their Metabolites in Human Urine Without Hydrolysis

Instrument Type: LCMSMS

Analysis of opiate and opioid metabolites in urine is most often done with a hydrolysis step that make total sample preparation time up to 24 hours. The method described here eliminates the hydrolysis step by analyzing the conjugated metabolites intact using a Thermo Scientific Prelude SPLC system for sample preparation and a Thermo Scientific TSQ Endura triple quadrupole mass spectrometer for analysis. For forensic use only.

Top Down Milk Protein Identification and Relative Quantification by Q Exactive Mass Spectrometer

Instrument Type: LCMSMS

Whey proteins, most notably alpha-lactalbumin and beta-lactoglobulin, are currently being investigated for their potential positive health benefits. Whey proteins are a substantial percentage of human milk (~60%) while such proteins are less abundant in cow milk (~20%). We describe a method for the identification of the various forms of caseins and whey proteins as a first step and then intact protein-based method for determination of the whey vs. casein protein ratio.

Fast Online Desalting of mAbs Using a Reversed-Phase Desalting Cartridge for LC-MS Analysis

Instrument Type: LCMS

This collateral describes a LCMS method for high throughput intact antibody characterization, capable of delivering the intact mass and the relative glycoform abundance of a mAb within 5 minutes. The desalting method optimization was conducted on a 2.1 × 10 mm Thermo Scientific™ MSPac™ DS-10 cartridge using conventional water/acetonitrile-based separations on the Thermo Scientific™ Vanquish™ UHPLC system and MS detection using the Thermo Scientific™ Q Exactive™ Plus Hybrid Quadrupole-Orbitrap™ mass spectrometer.
