Determination of carbohydrates in peanuts by HPAE-PAD

Peanuts contain the digestible carbohydrates sucrose, fructose, and glucose, but they also often include non-digestible raffinose, stachyose, and verbascose in varying quantities that pass through the human digestive tract unchanged and are subject to fermentation in the lower gut. For determinations of carbohydrates in food, a method for carbohydrates in peanuts and peanut butters with minimal sample preparation and without requiring pre- or post-column analyte derivatization. The method uses high-performance anion-exchange chromatography with pulsed amperometry detection (HPAE-PAD).

Biomass burning is a major source of particulate matter present in atmospheric aerosol. Anhydrosugars, sugar alcohols, monosaccharides, and disaccharides are among the major classes of water-soluble organic compounds (WSOC) in atmospheric aerosols. Saccharides have been proposed as markers to evaluate the contribution of these sources to atmospheric aerosols. Here, we developed an improved single column HPAE-PAD method to determine anhydrosugars, simple sugars, and sugar alcohols present in atmospheric aerosol samples using CarboPac PA300 column.

AN000170: A Fast HPAE-PAD Method for Determination of Carbohydrates and Glycols in Pharmaceutical Formulations

This application note describes a convenient, fast, and sensitive method that uses a Thermo Scientific™ Dionex™ CarboPac™ PA300 column and electrolytic eluent generation. The Dionex CarboPac PA300-4μm column is packed with a new 4 μm particle size supermacroporous resin to achieve high efficiency and high-resolution separations. Using this column, a 16.8 min method that resolved six analytes, including glycol and other carbohydrates, was designed. Results for method linearity, robustness, and accuracy for glycol and carbohydrate quantification in OTC samples are discussed here.
