Increased speed and sample throughput of opioid analysis from human urine using micro-elution solid phase extraction

To describe a fast and reproducible optimization procedure for the isolation of opioids from human urine utilizing solid phase extraction (SPE). Subsequent analysis was carried out with liquid chromatography separation coupled to triple quadrupole mass spectrometry detection (LC-MS/MS).

Rapid Analysis of 25-Hydroxyvitamin D3, epi-25-Hydroxyvitamin D3, and 25-Hydroxyvitamin D2 in Plasma for Clinical Research Using High-Resolution Mass Spectrometry

Chromatographic separation is required for simultaneous analysis of 25-hydroxyvitamin D3 (25OHD3) and its epimeric form 3-epi-25-hydroxyvitamin D3 (epi-25OHD3) in a liquid chromatography-mass spectrometry (LC-MS) method because both compounds have the same molecular formulas and the same fragmentation spectra. We developed an LC method that provides baseline separation between the two isobaric compounds. We coupled this chromatographic method to an MS method, which collects high-resolution MS/MS spectra for compound identification and quantitation.

This work demonstrates the use of Thermo Scientific SOLAμ SPE product to enhance sample pre-concentration prior to analysis. Additional benefits include reduced workflow and improved stability for analytes susceptible to loss or degradation during evaporation and reconstitution. The use of a Thermo Scientific Accucore HPLC column provided fast and efficient separation without the need for an ultra high pressure system.
