Separation of Fc and Fab Fragments from Papain Digested Denosumab sample by Reverse Phase Chromatography

The Thermo Scientific Dionex UltiMate 3000 LC Bio-LC system is applied for the separation of Fc and Fab Fragments from papain digestion of Denosumab sample by reverse phase chromatography. The separation was performed on Thermo Scientific MabPac RP column with a UV detection of 280 nm. With a run time of 15 minutes, the method allows easy separation of Fab and Fc fragments from papin digested Denosumab sample.

Long-term stable peptide mapping using the Vanquish UHPLCsystem

Peptide mapping is one of the major analyses in biotherapeutics characterization. Here we show peptide mapping with the Thermo Scientific Vanquish UHPLC system. An efficient and reliable separation of peptides obtained from a bovine serum albumin (BSA) digest is done on a Thermo Scientific Acclaim RSLC 120, C18, 2.2 µm Analytical column. Excellent retention time and area precisions as well as peak capacities were achieved over a time period of more than 80 injections.

In this work, a digestion of bovine serum albumin is resolved using the Thermo Scientific Acclaim RSLC C18 into 127 peaks with an average peak width of 0.27 minutes. For a 90 minute gradient, this works out to a theoretical peak capacity of >300.
