AN000472: HPAE-PAD Method for Determination of Hib Capsular Polysaccharide Content

This work describes an HPAE-PAD method for the determination of ribitol in Hib capsular polysaccharide acid hydrolysis samples. Separation of ribitol from TFA acid hydrolysis products was achieved under isocratic elution conditions using a Thermo Scientific™ Dionex™ CarboPac™ MA1 column. The method proposed here was validated for performance with respect to linearity, precision, sensitivity, and accuracy. The acid hydrolysis samples were also tested using a palladium hydrogen reference electrode.

AN72610: Determination of Carbohydrates in Algal Biofuel Samples (Part B: Long Method)

The current work describes two methods that use a Dionex CarboPac PA20-4µm column for determination key sugars present in acid hydrolyzed algal biomass. The long method separates the two uronic acids in addition to the twelve other sugars separated by the shorter method. When a detailed sugar composition calculation is desired, the long method can be used. The method was validated for precision, accuracy, and robustness during routine use.

The short method separates twelve common carbohydrate sugars in less than fifteen minutes. This method does not separate uronic acids, which are key components that are present in low concentrations. The method was validated for precision, accuracy, and robustness during routine use.

This Application Note describes the use of two different anion-exchange columns with amperometric detection to analyze simple sugars, sugar alcohols, alcohols, and glycols in yeast and bacterial fermentation broths. This record describes the CarboPac MA1 column method. The yeast Saccharomyces cerevisiae in Yeast Extract-Peptone-Dextrose (YPD) broth and the bacteria Escherichia coli (E. coli) in Luria-Bertani (LB) broth are common eukaryotic and prokaryotic fermentation systems, respectively.

TN137: Determination of Carbohydrates in Urine by Capillary HPAE-PAD

Certain carbohydrates such as mannitol, rhamnose, xylose, 3-O-methylglucose, and lactulose can be determined in urine and serum samples to evaluate disease states in animals and humans. These carbohydrates can be determined directly by HPAE-PAD, a well-established technique that eliminates the need for derivitization, in turn saving time and money. In this work, the separation and detection of the same carbohydrates of interest discussed in AN 1006 are demonstrated using a capillary Dionex CarboPac PA20 anion-exchange column on the ICS-4000 Dedicated Capillary HPIC system.
