AN163: Determination of Protein Concentrations Using AAA-Direct.

In this application note, we describe a method to determine total protein using AAA-Direct. We used this method to participate in an Association of Biomolecular Resource Facilities (ABRF) Amino Acid Analysis Research Group collaborative study that compared total protein determinations using AAA to dye-binding assays. We found that concentrations of all five protein solutions determined by AAA-Direct were equivalent to the determinations made by participants that used either pre- or postcolumn derivatization methods.

Improved analysis of API and counterions in Adderall

The Thermo Scientific UltiMate 3000 HPLC system is applied for the analysis of the API (active pharmaceutical ingredient) amphetamine and its counterions. The separation is performed using a Thermo Scientific Acclaim Trinity P1 column, with both UV and charged aerosol detection. Amphetamine and saccharin respond to UV detection; aspartate, saccharin and sulfate respond to charged-aerosol detection. Note that metal contamination will cause peak tailing, and before use, the column should be cleaned with a chelating agent at pH 4.

The separation is performed using a Thermo Scientific Acclaim Trinity P2 column, with charged aerosol detection. The Thermo Scientific Acclaim Trinity P2 column offers ideal selectivity for various cations and anions including organic acids. In this case, all label claimed minerals and their counterions are confirmed.
