CAN119: Determination of Human Milk Oligosaccharides in Human Breast Milk by HPAE-PAD with On-Line Sample Cleanup

In this study, a method for quantification of six human milk oligosaccharides, namely 2'-fucosyllactose, 3`-fucosyllactose, 3'-sialyllactose, 6'-sialyllactose, lacto-N-tetraose, and lacto-N-neotetraose in breast milk has been developed and validated. The method is based on the HPAE-PAD technique for carbohydrate determinations without sample derivatization. With an on-line sample cleanup using a polymeric reversed-phase column, a “dilute-and-shoot” sample preparation scheme was applied, which significantly reduces sample handling time and therefore increases sample throughput.

TN26: Determination of Cr(VI) in Water, Wastewater, and Solid Waste Extracts

The method presented here overcomes analyte interference by separating the two chromium using a detection method specific for Cr(VI) that is capable of handling the high-ionic-strength sample matrices generated in many leaching, and digestion procedures. Cr(VI) is separated as the divalent anion on the IonPac AS7 column using ammonium sulfate, ammonium hydroxide eluent. After the separation, Cr(VI) reacts with the color reagent diphenylcarbohydrazide (DPC) and detected by UV absorbance at 520 nm. This method is consistent with U.S. EPA Method 218.6. AB105: Anions and Organic Acids in Wood Extracts

This application demonstrates the analysis of inorganic anions and anions of organic acids in waste liquor from wood extracts. The sample matrix and the contaminant were removed from the components of interest using an IonPac NG1 column as an additional guard column. This application requires an autosampler equipped with 10-port valve. With this configuration, IonPac NG1 column is placed in line with the analytical column, while a second column rinsed in counter flow direction with acetonitrile/water, followed by a conditioning step using sodium hydroxide.

AB165: Determination of p-Toluenesulfonic Acid in Water-Insoluble Drugs

Here, the convenience and accuracy of a Reagent-Free IC (RFIC) approach is applied to the determination of p-toluenesulfonic acid in water-insoluble drugs using an on-line matrix-elimination as described in Application Note 220, albeit with different columns and a different system configuration. (Note- The method discussed here requires Thermo Scientific Dionex AS-AP Autosampler with optional valves (one 6-port valve and one 10-port valve for sequential injection).
