AN363: Using Accelerated Solvent Extraction in Alternative Fuel Research

The Accelerated Solvent Extraction (ASE) provides a fast and efficient way to extract various biomass samples for sugar analysis to determine sample viability for alcohol production. This application note describes methods for the extraction of sugars from biomass samples using acidified solvents (in-cell hydrolysis) and extracting pre-hydrolyzed samples.

AN1161: Improved Method for Determination of Biofuel Sugars by HPAE-PAD

The current work updates the system used for the carbohydrate analysis in AN1089. The new system combines flexibility and ease-of-use with high sensitivity and selectivity, bringing a higher level of convenience and cost effectiveness to simple sugar analysis. The method proposed here separates eight common carbohydrate sugars in less than eight minutes. The shorter run time results in better process economics. Using this method, carbohydrates present in 10 individual biofuel samples were quantified.

HPAE-PAD is widely used for determination of carbohydrates by measuring the electrical current generated by their oxidation at the surface of a gold electrode. Each amperometric detection cell includes an amperometric working electrode exposed to a sample flow-through channel, typically enclosed by a plastic gasket held in place by compression. Here, the sensitivity and linearity of six carbohydrates have been evaluated using 2, 15, and 62 mil gaskets under typical chromatographic conditions used for Thermo Scientific™ Dionex™ CarboPac™ SA10 column with and without postcolum addition.
