In this paper, a simple and efficient HPLC method was established by using Vanquish Core high performance liquid chromatography (Vanquish Core) combined with general quality detector-Charged Aerosol Detection(CAD). It can meet the needs of simultaneous determination of Poloxam 188 and Tween 80 in adeno-associated virus (AAV) vector preparation. The method has good stability and high sensitivity. The wide linear range is suitable for the determination of different concentrations of samples, and can be used as an effective means for the control of excipient residues in drugs such as Poloxam 188
Tween 80, or polysorbate 80, is a complex non-ionic surfactant which is quantified using a fast gradient HPLC method on a C1 column. The universal, mass-based Corona charged aerosol detector is used for quantitation of this polymeric analyte. Baseline subtraction is used to eliminate baseline effects. Limit of quantitation is 78 ng on column (o.c.), based on a peak area relative standard deviation (RSD) of 15%. The dynamic range was 78 - 5000 ng o.c. with R^2 of 0.9997 and a calibration RSD of 2.8%. This method is designed for a fast quantification of Tween 80 as a raw material.
